I came across this article on E! Online and I haven't seen this posted or talked about in the Dean & Jo community, maybe DJ fans are aware or not but I this comment disappointed me a bit.

Sex, Monsters and Full-Frontal Nudity: Supernatural Boss Spills! By Kristin Veitch Jan 11, 2007 3:26 AM
And how 'bout some lovin' for our boys? Is there something brewing between Jo and Dean? Eric Kripke: No. [Laughs.] That's the short answer. Um, I love the character of Jo, and I love Alona [Tal]. I think she's done such a great job. But I think she's sort of been established as more of a sister character to the boys, and it works well. It's a good dynamic, so yeah, no sex for Jo and Dean!......
I know it's from January but it also seems that maybe DJ could still happen with all the shipper fandom doing their support.

Yeah... To be honest, I'm was never at all crushed to hear "no sex for Jo and Dean," but I was and am sad to hear Kripke say that she will be more of a sister. What's worse, in one interview he described a romantic relationship between Dean and Jo as "trying to fit a square peg in a round hole."
Now, though it grieves me to say so, I can understand why such comments would be made in the context of No Exit. I like that episode, but I admit to be dissapointed in many areas with the manner in which Jo was written. Don't get me wrong - there are a lot of scenes Jo had in that episode which made me love her even more, but some scenes between Jo and Dean were very sister/brother-esque. For example, Dean lectured Jo quite frequently (telling her she oughtn't lie to her mother; telling her she should appreciate her options, etc., etc. The script indicated a brother/sister relationship and that carried over into their chemistry. I am confident that if Dean and Jo were given a script indicating a romantic relationship they would positively sizzle!

I don't know, I never felt much OF a sibling chemistry between Jo and Dean. I never saw a sexual chemistry either, but I thought that they had their very own chemistry... Jensen's chemistry with women is generally *sexual* or *sibling*, but the way Kripke created Dean and Jo, with all the mirrors, I feel a chemistry that potentially could develop into a romantic one.
What No Exit showed me was two people who, beyond the bickering and the lecturing, could understand each other. When I say that, I don't mean "understand" as a light word. Dean likes chicks but he's never clicked with with any of those. He was touched by Andrea and Layla and he loved Cassie at one point, but he can understand Jo. He can understand the fire in her eyes when she talks about hunting and he can understand the coldness in her tone when it comes to her father.
That's what No Exit was about to me. Not romance, not sex, not sibling chemistry. Just plain understanding.
And to me, that particular level of understanding shows in their chemistry. And I believe that from this point, whatever way their relationship goes, both actors could reach the kind of chemistry to go with their relationship. I can see them going either sibling, either romantic, either sexual. That's up to them and Kripke.

^you said it so well. In No Exit, Dean and Jo connected on a different level that Dean could never connect to with any other girl. the fact that there is a level of understanding there says alot. that doesn't necessarily imply that there might be some romantic undertones but it doesn't negate it either.
i loved the fact that Dean was able to open up to Jo about his past memory with his father and that Jo was able to do the same. he's human and all humans long for understanding. But there's just a few people who are ever able to "get" Dean and Jo is one of em. (well at least i think so )

I just can't agree more. Besides, Dean needs a hard headed woman in his life... not just another stupid-dollgirl to play one night. Dean/Jo has big potencial... I just can't believe Kripke's still have the idea of a sister for the boys why Jo???? There are so many actresses out there... why Alona, why Jo??? why????? Anyways, I'm still waiting for that specific moment of Dean and Jo making out hahahah or something like that in Supernatural.

Oh, Amen! I could not have put that sentiment any better myself. As I said, there were some scenes to No Exit that I thought ought to have been written differently, or perhaps in which Alona ought to have been directed differently, but when everything is said and done I think your'e absolutely right.
